Acquiring a population of cells from a single cell

  • Simple in unicellular organisms, such as yeast and bacteria
    • Only requires the inoculation of the appropriate medium
  • Cloning rings used to clone distinct lineages of cell lines
    • Single-cell suspension of cells exposed to a mutagenic agent is plated at high dilution to create isolated colonies
    • Sterile cloning rings which have been dipped in grease are placed over an individual colony at an early growth stage
    • Trypsin, an agent that breaks down proteins, is added
    • Cloned cells are collected from the ring and transferred to a vessels for further growth
