The biological amplification of a specific DNA sequence through miotic division of a host cell

  • Used in a variety of biological experiments and practical applications, such as protein production
  • Four steps:
    • Fragmentation: DNA of interest is isolated to provide revelant DNA of the proper size
    • Ligation: Vector is linearized by enzymes and incubated with fragment of interest with the enzyme DNA ligase
    • Transfected: Vector with insert of interest is transfected into cells
    • Screening/Selection: Transfected cells are cultured
  • Term to know: amplification- an increase in the number of copies of a specific DNA fragment
